On the path to age-defying therapies

ScienceDaily (Mar. 29, 2012) ? One of the secrets to a longer, healthier life is simply to eat less. When subjected to calorie restriction (CR), typically defined as a 20-40% reduction in caloric intake with corresponding maintenance of proper nutrition, animals in labs not only live longer, but also have improved insulin sensitivity and glucose tolerance, both of which decline during aging.

Yet, for all of its benefits, CR's restricted diet is a stumbling block for most Americans. If only we had a drug that could do the same thing.

Well, we do, sort of. The drug rapamycin, which is used for immunosuppression in organ transplantations, mimics the longevity effects of CR and may tap into the same cellular pathway as CR. Unlike CR, however, rapamycin actually impairs glucose tolerance and insulin sensitivity, two hallmarks of diabetes. Clearly, rapamycin is doing something CR is not.

To understand better rapamycin's benefits and risks, researchers from the lab of Whitehead Institute Member David Sabatini and Joseph Baur, assistant professor of Physiology, at the University of Pennsylvania's Perelman School of Medicine, have discovered precisely how rapamycin is behaving at the cellular level. Their intriguing results are published this week in the journal Science.

"We know that despite its adverse effects, rapamycin still prolongs lifespan, so there's a potential that we could make it better by just having lifespan affected and not induce the adverse effects," says Sabatini, who is a professor of biology at MIT and a Howard Hughes Medical Institute (HHMI) investigator. "The data in this paper suggest that it's possible."

Rapamycin, which is also called sirolimus and marketed in the United States as Rapamune, is a known inhibitor of the mechanistic target of rapamycin complex 1 (mTORC1), a protein complex that regulates many cellular processes linked to growth and differentiation. mTORC1 is part of a cellular signaling pathway, called mTOR, which responds to nutrients and growth factors. Mechanistic target of rapamycin complex 2 (mTORC2) is also part of the mTOR pathway and regulates insulin signaling.

Rapamycin has generally been thought to target primarily mTORC1. But work by Dudley Lamming and Lan Ye, co-authors of the Science paper and postdoctoral fellows in the Sabatini and Baur labs respectively, indicates that in mice, rapamycin also inhibits mTORC2, thereby reducing insulin sensitivity.

To see if rapamycin's positive effects on lifespan effects could be separated from its negative metabolic effects, Lamming and Ye bred mice whose mTORC1 activity was partially inhibited but whose mTORC2 activity remained largely intact. The females of this mouse population lived longer than control mice while maintaining normal insulin sensitivity.

"This shows that disrupting mTORC1 alone is capable of extending lifespan, if you can find a way do that," says Lamming.

For Baur, the experiments' results indicate that there is a possibility of identifying a better anti-aging drug than rapamycin.

"Our work highlights the potential utility of molecules that target mTORC1 specifically and suggests there is hope that by targeting this pathway, you could really get something that ameloriates age-related diseases without causing more problems than it solves," says Baur. "If you're taking an anti-aging drug as a preventive measure, you probably don't want to pay the price of diabetes."

A giant mass of sand large enough to bury all of Manhattan under dunes more than 50 stories tall apparently erupted from the floor of the North Sea hundreds of thousands of years ago, the largest such body of sand ever found in the world, researchers say.

Scientists used 3-D seismic data and samples drilled from the ocean bottom during fossil fuel exploration to analyze a huge body of sand that lies over an oil field in the North Sea. Their research suggested it was "extrusive," spewed out from fissures in the seafloor likely over the course of years sometime between 400,000 and 2.6 million years ago.

At 2.4 cubic miles (10 cubic kilometers) in size, this is apparently the largest extrusive body of sand ever discovered. This is big enough to bury Manhattan's 23 square miles (60 square kilometers) under 525 feet (160 meters) of sand, or the whole of London's 610 square miles (1,579 sq km) under 20 feet (6 m) of sand.

"Most exciting is discovering things for the first time and realizing that you have discovered something new," researcher Helge Loseth, a geologist at the Statoil Research Center in Trondheim, Norway, told OurAmazingPlanet.

Currently, the sand covers an area of more than 100 square miles (260 sq km) and is up to 410 feet (125 m) thick. It lies buried under about 2,300 feet (700 m) of sediment that accumulated on top of it after its deposition, with at least another 1,640 feet (500 m) of the North Sea above that.

Loseth explained that under the surface of the seafloor, porous sands are covered with impermeable clays. The water pressure between the sand grains can increase enough to fracture the overlying clays, and this pressurized sandy water will then erupt upward.

"This is similar to when you increase the water pressure in one end of your garden hose by turning the tap," Loseth said.

Aside from being interesting for its size, this body of sand might have economic value as well. The pores between sand grains are normally filled with water, but hydrocarbons such as oil or gas can migrate into these pores instead.

"Such big sands are so large that they can contain economically interesting volumes of hydrocarbons," Loseth said. "Today, at the depth they are placed in the North Sea, they may also be interesting for carbon dioxide storage."

Loseth and his colleagues Nuno Rodrigues and Peter Cobbold detailed their findings online March 19 in the journal Geology.

egl: Review of an Usagi Youhinten Purchase

Hello egl!

Recently I placed an order with the Japanese consignment shop Usagi Youhinten and since there doesn't seem to be too many reviews on the shop, I've decided to write one :)

A couple of weeks ago I spotted BtSSB's Heart Flocky Ribbon JSK in black on Usagi Youhinten's website, and fell in love with it immediately - it was time for some more internet shopping! I ordered via email, desperately hoping that it would not be sold before I got a reply because I was hesitant with messing up some details if I used the shopping cart in Japanese.

19 March -?Sent an initial email asking about sizing and shipping costs of the JSK. A staff member replied within a couple of hours with helpful answers and a shipping quote with three options: EMS, small packet with registered airmail, Economy (SAL). All three are insured and have tracking, and the only difference is their shipping times~

20 March -?Confirmed my purchase of the JSK, and added a headbow to my order. An hour later I decided to cancel the headbow (derp) and sent another email to which the staff replied quickly and courteously, changing my order without any problems.
I was invoiced and paid through PayPal in the same evening, and confirmation of receiving my payment was quick.

In total my purchase cost:
8800 yen JSK price
1680 yen Economy shipping price (total weight was about 570g with a cardboard box)

21 March -?I received an email to say that my package had been sent. I chose Economy/SAL, which was the cheapest option but the slowest (quoted at 2 weeks delivery). I had to ask for the tracking number, but received it quickly after requesting it.

28 March - I was very surprised to come home to my parcel only a week after it was shipped!

The box that arrived! It was very sturdy and barely dented. It was secured with ten million strips of tape that took some time to get off.?

The JSK was wrapped folded neatly inside?two?sturdy plastic bags. They even had some cute heart-printed tape on the back, and a sticker with my name on it :)

Aaaand the JSK! It was listed as?A-?on the website's description so I'm guessing it's been worn a couple of times. This definately did not show, however and it honestly looks brand-spanking new! There was a faint factory-like smell, but I don't fault that because it's probably been in storage for a while.

Closeup of the bottom hem: The flocky hearts are so cute! You can see a few specks of dust, but with the fabric of the JSK, there was bound to be some unless it was kept constantly covered. A quick brush down removed most of the white flecks/dust.

Egyptian parliament names constitution panel

CAIRO (AP) ? Egypt's Islamists looked poised Saturday to gain control of a key lever of power that will help steer the country's political future as parliament selected a panel to draw up the country's new constitution. Liberal lawmakers denounced the process as a "farce" and walked out in protest.

The constitution, which will be written by the 100-member committee, will determine the balance of power between Egypt's previously all-powerful president and parliament, and define the country's future identity, including the role of religion and minority rights. With so much at stake, the question of who should sit on the panel has sparked fierce debate in Egypt.

Many secular and liberal Egyptians fear that the Islamist parties that dominate parliament will pack the panel with their supporters and ignore minority concerns. Those fears have spiked over the past week after parliament decided to allocate half of the panel's seats to its own members, and a leading Islamist deputy said the country's most prominent democracy advocate, Mohamed ElBaradei, would likely not be included.

With vote-counting still going on late Saturday night, Egypt's state-run MENA news agency posted a list of names of those it said were gaining support among lawmakers.

The list included more than 50 Islamists, as well as six women and six Christians in addition to some 30 non-Islamists, including professionals, judges, political scientists, poets, and others. Some of the names were not widely known to the public and their ideological affiliation was not immediately clear.

During the heated joint session of parliament to pick the panel, lawmakers from the liberal Egyptian Bloc, which holds 9 percent of the lower house's seats, walked out of the proceedings en masse, accusing the fundamentalist Muslim Brotherhood ? Egypt's most powerful political group ? of trying to dominate the selection.

Egyptian Bloc lawmaker Emad Gad called the session a "farce" and said the Islamist parties "have already decided on the names and voting is only meant to lend legitimacy to the process."

Saad Emara, a Muslim Brotherhood lawmaker, accused liberals of "making a fuss," and said millions of Egyptian voters elected Islamist lawmakers so that their views would be represented on the constitutional committee.

Egypt's ruling military council last year issued an interim constitution that gives elected members of the parliament's two houses the right to select those who will draft the new constitution. The old 1971 constitution was suspended after the uprising that ousted longtime ruler Hosni Mubarak.

The new constitution will be put to a vote in a national referendum. However, the ruling military council left the guidelines for the process vague enough to spark a sharp debate over who should be on the panel.

Egypt's Islamist groups, including both the Brotherhood and the ultraconservative Salafis, make up nearly three-quarters of parliament after sweeping the vote in the first post-revolution elections that began in November.

They passed a vote last week to appoint 50 of the panel members from among lawmakers in parliament, while the rest will be drawn from broader society.

Liberals, among whom are youth groups and secular parties that led the uprising but performed poorly in elections, say that a permanent constitution should not be written solely by the victors of a single election.

They argue that the constitutional process should include a wide range of members from the country's different ideological currents, professional syndicates and unions, women, and members of the Christian minority. They say that parliament's decision to have its members dominate the process violates earlier Brotherhood pledges to draft the charter by "consensus" and fear it represents a capitulation to the hardline Salafis.

As the Brotherhood squares off with the liberals and secularists in parliament, it's also engaged in a standoff with the ruling military council.

The 83-year-old Brotherhood, which was officially banned under Mubarak, said in a statement Saturday that it has received threats from the military-backed government that it could move to have parliament dissolved in response to the Brotherhood's pressing the army to sack the current Cabinet.

Brotherhood deputy leader Rashad Bayoumi said Prime Minister Kamal el-Ganzouri threatened to use a court ruling to dissolve parliament. Bayoumi declined to say when the threat was made.

The new constitution is expected to curb presidential powers and give parliament more authority, a drastic change to Egypt's political system.

Another key concern is the role of Islamic Shariah law, which is subject to a wide variety of interpretation.

The old constitution says Shariah is the "main source of legislation," but many in the hardline Salafi bloc that makes up nearly a quarter of parliament's members take a literalist and uncompromising stance on Shariah and want it strictly enforced.

Another divisive issue is the role of the military and the future of the country's military rulers. The ruling generals want assurances they won't lose their political clout and that parliament will have no say over the military's budget.

Parliamentary speaker Saad el-Katatni, a member of the Muslim Brotherhood, promised this month that the new constitution should not be written by "the majority," but instead by "consensus and partnership."

That pledge however has been called into question by the exclusion of ElBaradei, whom Brotherhood parliamentarian Mohammed el-Beltagi said on his Facebook page Friday would normally be included "only if he didn't oppose the current road map" for drafting the document.

ElBaradei had criticized the parliament ? the product of the first open elections after decades of dictatorship ? as not fully representative, and the process of drafting the constitution as rushed.

With drums and chants, youth activists rallied outside parliament against Islamists and the military for what they see as sabotaging the revolution.

"No Salafis, no Brotherhood. The constitution is for all Egyptians," they chanted.

Employers and colleges that demand Facebook passwords or other access to social network profiles are doing more than violating the First Amendment, as advocates argue. They're also violating Facebook's terms of service.

Almost three weeks after Bob Sullivan of msnbc.com's Red Tape Chronicles first broke the story of government agencies and colleges demanding inappropriate access to Facebook accounts, both the social network and a U.S. senator are calling shenanigans on this violation of privacy, which is also a potential liability problem for the inquiring institutions.

On Friday,?Erin Egan, Facebook's chief privacy officer on policy, was direct?and referenced the possibility of legal action.

The practice of requesting Facebook passwords and other access to user profiles "undermines the privacy expectations and the security of both the user and the user's friends," Egan said in a statement on the social network's privacy page. "It also potentially exposes the employer who seeks this access to unanticipated legal liability."?

As an example, Egan?posited, "if an employer sees on Facebook that someone is a member of a protected group (e.g. over a certain age, etc.) that employer may open themselves up to claims of discrimination if they don't hire that person."

Egan went on to say,?it is now a "violation of Facebook's Statement of Rights and Responsibilities to share or solicit a Facebook password."

On Wednesday,?Sen. Richard Blumenthal D-Conn. told Politico?he's writing a bill that would outlaw the practice as well:

The senator cited longstanding federal prohibitions against administering polygraph tests to potential employees as a rationale for a new law.
Even for prospective employees who voluntarily surrendered account access to improve their chances of landing a job, Blumenthal said there remain concerns. "The coercive element of the request," he said, "really makes it less than voluntary."

ACLU attorney Catherine Crump had stronger words for the practice. "People are entitled to their private lives,"?she said in a statement. "You'd be appalled if your employer insisted on opening up your postal mail to see if there was anything of interest inside. It's equally out of bounds for an employer to go on a fishing expedition through a person's private social media account."

Here are some of the egregious examples of Facebook privacy violations,?first reported by Sullivan on March 6:

In Maryland, job seekers applying to the state's Department of Corrections have been asked during interviews to log into their accounts and let an interviewer watch while the potential employee clicks through wall posts, friends, photos and anything else that might be found behind the privacy wall.
Previously, applicants were asked to surrender their user name and password, but a complaint from the ACLU stopped that practice last year. While submitting to a Facebook review is voluntary, virtually all applicants agree to it out of a desire to score well in the interview, according Maryland ACLU legislative director Melissa Coretz Goemann.
Student-athletes in colleges around the country also are finding out they can no longer maintain privacy in Facebook communications because schools are requiring them to "friend" a coach or compliance officer, giving that person access to their ?friends-only? posts. Schools are also turning to social media monitoring companies with names like UDilligence and Varsity Monitor for software packages that automate the task. The programs offer a "reputation scoreboard" to coaches and send "threat level" warnings about individual athletes to compliance officers.

As well as likely violating the First Amendment, such practices also herald legal problems for the institutions, noted Bradley Shear, a Washington D.C.-lawyer who first brought these cases to the attention of Maryland polticians and the ACLU. As an example of liability, he told Sullivan:

"What if the University of Virginia had been monitoring accounts in the Yeardley Love case and missed signals that something was going to happen?? he said, referring to a notorious campus murder. ?What about the liability the school might have?"

Following scrutiny by the ACLU, Maryland state legislators proposed two separate bills aimed at banning social media access by schools and potential employers, Sullivan reported. At the time of the original story, a Facebook spokesperson wouldn't specifically address the state legislation except to say that many of these school and employer policies appear to violate the site's terms.

Though it was more than two weeks in coming, a?definitive?statement and action from Facebook against such privacy violations is in the social network's best interest, more so than the interests of its users. Allowing the practice ?to continue means more organizations will feel free to make password demands, making users feel increasingly less comfortable sharing their personal information via Facebook. Less sharing on Facebook means less personal info for targeted marketing, which is the social network's bread and butter.?

As Facebook continues to make us more comfortable with less and less privacy, this may be one of the few cases where everybody benefits from the social network's pro-sharing business model.?
